Abstract:
ThemechanisticunderstandingofmetalhyperaccumulationhasbenefittedimmenselyfromtheuseofmoleculargeneticstoolsdevelopedforArabidopsisthaliana.TherevolutioninDNAsequencingwillenableevengreaterstridesinthenearfuture,thistimenotrestrictedtothefamilyBrassicaceae.Referencegenomesarewithinreachformanyecologicallyinterestingspeciesincludingheterozygousoutbreeders.TheywillallowdeepRNA-seqtranscriptomestudiesandthere-sequencingofcontrastingindividualstounravelthegeneticbasisofphenotypicvariation.Cell-typespecifictranscriptomeanalyses,whichwillbeessentialforthedissectionofmetaltranslocationpathwaysinhyperaccumulators,canbeachievedthroughthecombinationofRNA-seqandtranslatomeapproaches.Affordablehigh-resolutiongenotypingofmanyindividualsenablestheelucidationofquantitativetraitlociinintra-andinterspecificcrossesaswellasthroughgenome-wideassociationmappingacrosslargepanelsofaccessions.Furthermore,genome-widescanshavethepowertodetectlociunderrecentselection.Togethertheseapproacheswillleadtoadetailedunderstandingoftheevolutionarypathtowardstheemergenceofhyperaccumulationtraits.
